Within this study, essential metabolic pathways and candidate genes involved in male sexual improvement had been selected from M. nipponense. The methodology involved performing long-read and subsequent generation transcriptome sequencing of genes in the androgenic gland following eyestalk ablation. qPCR evaluation revealed that the mRNA expression of Mn-IAG was drastically increased right after ablation of each the single-side (SS) and double-side (DS) eyestalk, compared with the handle group (CG). The long-read transcriptome generated 49,840 non-redundant transcripts. A total of 1319, 2092 and 4351 differentially expressed genes (DEGs) had been identified between CG versus SS, SS versus DS and CG versus DS, respectively. These data indicated that ablation in the double-sided eyestalk played stronger regulatory roles than the single-side ablation on male sexual improvement in M. nipponense. The oriental river prawn, Macrobrachium nipponense (Crustacea; Decapoda; Palaemonidae), is extensively distributed in China as well as other Asian countries1. This species is NOP Receptor/ORL1 list commercially important with an annual aquaculture production of 205,010 tons in 20164. Comparable to other Macrobrachium species, male prawns develop more quickly and reach a larger size at harvest time2. Therefore, male prawns are preferred in the M. nipponense aquaculture business.
