Scientific Background
MAPK4 is a protein-serine/threonine kinase that phosphorylates microtubule-associated protein 2 (MAP2) and MAPKAPK5. The precise role of the complex formed with MAPKAPK5 is still unclear, but the complex follows a complex set of phosphorylation events: upon interaction with atypical MAPKAPK5, ERK4/MAPK4 is phosphorylated at Ser-186 and then mediates phosphorylation and activation of MAPKAPK5, which in turn phosphorylates ERK4/MAPK4. MAPK4 may promote entry in the cell cycle.

Scientific Background
MAP3K8 is a protein serine/threonine kinase that can activate both MAPK and JNKK pathways. It is required for TLR4 activation of the MEK/ERK pathway. It can activate NF-kB1 by stimulating the proteolysis of its inhibitor, p105, and promote the production of TNF-alpha and IL-2 during T-cell activation. It has been shown to mediate Raf-inhibiting drug resistance in BRAF(V600E) cell lines. MAP3K8 may also play a role in the cell cycle.

Scientific Background
MAP3K8 is a protein serine/threonine kinase that can activate both MAPK and JNKK pathways. It is required for TLR4 activation of the MEK/ERK pathway. It can activate NF-kB1 by stimulating the proteolysis of its inhibitor, p105, and promote the production of TNF-alpha and IL-2 during T-cell activation. It has been shown to mediate Raf-inhibiting drug resistance in BRAF(V600E) cell lines. MAP3K8 may also play a role in the cell cycle.

Microtubule-associated protein 2 (MAP2) is a protein that in humans is encoded by the MAP2 gene. MAP2 is essential for the development and maitenance of neuronal morphology. The proteins of this family are thought to be involved in microtubule assembly, which is an essential step in neuritogenesis. MAP2 serves to stabilize microtubules (MT) growth by crosslinking MT with intermediate filaments and other MTs.
