Scientific Background
Antigen KI-67 also known as Ki-67 or MKI67 is a protein that in humans is encoded by the MKI67 gene. Antigen KI-67 is a nuclear protein that is associated with and may be necessary for cellular proliferation. Furthermore, it is associated with ribosomal RNA transcription. Inactivation of antigen KI-67 leads to inhibition of ribosomal RNA synthesis. Due to its association with cell divison process, Ki-67 is routinely used as cellular proliferation marker of solid tumors as well as certain hematological malignancies, and a correlation has been demonstrated between Ki-67 index and the histopathological grade of cancers.

Scientific Background
The endoplasmic reticulum is part of a protein sorting pathway, or in essence, the transportation system of the eukaryotic cell. The majority of endoplasmic reticulum resident proteins are retained in the endoplasmic reticulum through a retention motif. This motif is composed of four amino acids at the C-terminal end of the protein sequence. The most common retention sequence is KDEL (lys-asp-glu-leu). However, variation on KDEL does occur and other sequences can also give rise to endoplasmic reticulum retention (6). There are three KDEL receptors in mammalian cells, all have a very high degree of sequence identity; and all are located within the cis-Golgi and its intermediate compartments (4).
In terms of function, KDEL receptors interact with GAP (GTPase-activating protein) of ARF1, which is involved in COPI dependent vesicle transport, and the KDEL receptor may also be responsible for the recruitment of this ARF1 to membranes which can then aid in the regulation of vesicle budding (3). It is also important to note that the KDEL receptor exhibits extensive sequence identity o yeast protein Erd2p, which is a receptor for the yeast ER retention signal (4, 5).

Scientific Background
The endoplasmic reticulum is part of a protein sorting pathway, or in essence, the transportation system of the eukaryotic cell. The majority of endoplasmic reticulum resident proteins are retained in the endoplasmic reticulum through a retention motif. This motif is composed of four amino acids at the C-terminal end of the protein sequence. The most common retention sequence is KDEL (lys-asp-glu-leu). Grp78 and Grp94 and PDI all share the C-terminal KDEL sequence. The presence of carboxy-terminal KDEL appears to be necessary for ER retention and appears to be sufficient to reduce the secretion of proteins from the ER.

The protein encoded by this gene forms a potassium channel that is thought to play a critical role in the regulation of neuronal excitability (1), particularly in sensory cells of the cochlea (2). The current generated by this channel is inhibited by M1 muscarinic acetylcholine receptors and activated by retigabine, a novel anti-convulsant drug (3).

Scientific Background
Kv7.1 (KvLQT1) is a potassium channel protein coded for by the gene KCNQ1. Kv7.1 is present in the cell membranes of cardiac muscle tissue and in inner ear neurons (1) among other tissues. In the cardiac cells, Kv7.1 mediates the IKs (or slow delayed rectifying K+) current that contributes to the repolarization of the cell, terminating the cardiac action potential and thereby the heart's contraction (2, 3).

Scientific Background
KCC2 is a member of the cation-chloride cotransporter gene family (1). It acts as a K-Cl cotransporter. KCCs normally lower intracellular chloride concentrations below the electrochemical equilibrium potential and depending on the chemical concentration gradients of potassium and chloride, KCC2 can operate as a net efflux or influx pathway. It is proposed to act as the main chloride extruder to promote fast hyperpolarizing postsynaptic inhibition in the brain (2, 3). KCC2 is expressed at high levels in neurons throughout the nervous system and immunofluorescence shows that the protein is localized at inhibitory synapses of the spinal cord (4). Studies in mice have shown that KCC2 reduces GABA's inhibitory signaling, resulting in motor defects, epilepsy, and anxiety-like behavior.
