Scientific Background
Acetylcholine (ACh) is a common neurotransmitter for motoneurons, preganglionic autonomic neurons, postganglionic parasympathetic neurons, a variety of brain regions and some emerging neuron-like stem cells. The metabolism of Ach is relatively simple, involving only two enzymes: choline acetyltransferase (ChAT) for synthesis and acetylcholinesterase (AChE) for degradation. Further, acetylcholine has little function in neurons other than neurotransmission and seems to be neuron specific. It seems that only cholinergic neurons have significant amounts of ChAT making anti-choline acetyltransferase a useful specific marker. ChAT is a valuable marker for diseases associated with decreased cholinergic function such as Schizophrenia, Alzheimer disease and Down syndrome (1-3).

Scientific Background
Chloroquine diphosphate was originally developed as a treatment against malaria. It has since been implicated in cancer research as it inhibits cell growth and induces cell death in a number of cancer cell lines. Chloroquine also inhibits autophagy by specifically blocking acidification of the lysosomes which fuse with autophagosomes. This consequently prevents the degradation of materials.

Scientific Background
Chk2 is a protein kinase involved in cell cycle arrest in response to DNA damage. CHK2 is activated through phosphorylation by ATM and ATR at T68. Activated CHK2 phosphorylates and inhibits CDC25 a dual-specificity phosphatase required for progression through the cell cycle. CHK2 further causes cell cycle earrest through activation of a DNA repair protein BRCA1 and apoptosis factor PML. The role of CHK2 in the development of cancer is in a primarily tumour suppressor role through causing cell cycle arrest. It has been observed that inhibition of CHK2 through mutations in the regulatory FHA domain has been observed in low frequency in osteosarcomas and cancers of the ovary, lung and vulva. Another mutation, "1100delC" has been commonly found in breast cancers.

Scientific Background
Chk1 is a protein-serine/threonine kinase required for checkpoint-mediated cell cycle arrest. It also subsequently activates DNA repair (i.e. homologous recombination) by phosphorylating RAD51 or FANCE in response to the presence of damage or unreplicated DNA. It regulates cell cycle progression via several mechanisms: 1) phosphorylation of certain proteins such as CDC25A, CDC25B and CDC25C, promoting their degradation; 2) phosphorylation and activation of p53; 3) phosphorylation and inactivation of TLK1, which consequently prevents TLK1-dependent phosphorylation of chromatin assembly factor ASF1A; 4) phosphorylation of histones and regulating transcription of genes involved in cell-cycle progression regulation; 5) phosphorylation of RB1 to promote its interaction with E2F transcription factors and subsequent cell cycle arrest

Scientific Background
Chapsyn-110, is a part of the membrane-associated putative guanylate kinase (MAGUK) family. It binds directly to the NMDA receptor, and Shaker K+ channel subunits, and is 70-80% identical to PSD-95/SAP90 and SAP97 (1). It associated tightly with the postsynaptic density I brain, and mediates the clustering of both NMDA recpetors and K+ channels in heterologous cells. The encoded protein forms a heterodimer with PSD-95 that may interact at postsynaptic sites to form a multimeric scaffold for the clustering of receptors, ion channels, and associated signaling proteins (1, 2).

AP (Alkaline Phosphatase)
Properties:
- Broad enzymatic activity for phosphate esters of alcohols, amines, pyrophosphate, and phenols
- Commonly used to dephosphorylate the 5’-termini of DNA and RNA to prevent self-ligation
- Catalyzes the conversion of:
- Chromogenic substrates (e.g. pNPP, naphthol AS-TR phosphate, BCIP) into coloured products
- Fluorogenic substrates (e.g. 4-methylumbelliferyl phosphate) into fluorescent products
- Molecular weight: 140 kDa
- Applications: Western blot, immunohistochemistry, and ELISA
HRP (Horseradish peroxidase)
Properties:
- Enzymatic activity is used to amplify weak signals and increase visibility of a target
- Readily combines with hydrogen peroxide (H2O2) to form HRP-H2O2 complex which can oxidize various hydrogen donors
- Catalyzes the conversion of:
- Chromogenic substrates (e.g. TMB, DAB, ABTS) into coloured products
- Chemiluminescent substrates (e.g. luminol and isoluminol) into light emitting products via enhanced chemiluminescence (ECL)
- Fluorogenic substrates (e.g. tyramine, homovanillic acid, and 4-hydroxyphenyl acetic acid) into fluorescent products
- High turnover rate enables rapid generation of a strong signal
- 44 kDa glycoprotein
- Extinction coefficient: 100 (403 nm)
- Applications: Western blot, immunohistochemistry, and ELISA
Biotin
Properties:
Streptavidin
Properties:
- Homo-tetrameric protein purified from Streptomyces avidinii which binds four biotin molecules with extremely high affinity
- Molecular weight: 53 kDa
- Formula: C10H16N2O3S
- Applications: Western blot, immunohistochemistry, and ELISA
